Functional and Binding Studies of the Roles of Prothrombin and p 2 - Glycoprotein I in the Expression of Lupus Anticoagulant Activity

چکیده
We present functional and binding data relevant to the reported roles for prothrombin and /32-glycoprotein I (p2GPI) in the expression of lupus anticoagulant activity. In a purified system containing human prothrombin, Xa, Va, and a ratelimiting concentration of phosphatidylserine (PS)/phosphatidylcholine (PC) vesicles, the preliminary incubation of vesicles with protein A separated IgG preparations from 10 lupus anticoagulant plasmas, calcium, and prothrombin enhanced the inhibitory effect of all IgG preparations upon thrombin generation. Experiments in a purified factor X activation system provided supporting data that a similar preliminary incubation with prothrombin enhanced the inhibitory effect of many of the IgG preparations upon factor X activation. However, we could not obtain unequivocal evidence that prothrombin was an obligatory cofactor for lupus anticoagulant IgG to inhibit procoagulant phospholipid function, because lupus anticoagulant IgG separated by protein A chromatography contained traces of prothrombin. The binding of many IgG preparations to immobilized PS was enhanced by prothrombin when calcium ions were present. P2GPI enhanced binding of many of the IgG preparations to immobilized PS both in the presence and absence of calcium, yet

متن کاملA mechanism for the hypoprothrombinemia of the acquired hypoprothrombinemia-lupus anticoagulant syndrome.
Antibodies that bind prothrombin without neutralizing its coagulant activity were demonstrated in the plasma of two patients with the acquired hypoprothrombinemia-lupus anticoagulant syndrome. The first patient's plasma contained less than 1% prothrombin activity and no detectable prothrombin antigen. The second patient's plasma contained about 6% of both prothrombin activity and antigen. Neith...
متن کاملComparison of Serum Level of Antiphospholipid Antibodies and Homocysteine in Patients with Pemphigus Vulgaris and Healthy Subjects: A Case-Control Study
Introduction: Pemphigus vulgaris is an autoimmune blistering disease of the skin and mucous membrane. Antiphospholipid antibodies are a group of autoantibodies found in patients with autoimmune diseases. Due to the risk for thrombotic events in autoimmune diseases, we decided to evaluate the correlation between pemphigus vulgaris and predictive elements of atherothrombosis including antiphospho...
متن کاملAnti-prothrombin antibodies and the lupus anticoagulant.
The investigators have evaluated the frequency and manifestations of anti-prothrombin antibodies in patients with the lupus anticoagulant. Thirty-one of 42 patients with lupus anticoagulants associated with a variety of underlying conditions (74%) had evidence on crossed immunoelectrophoresis of anti-prothrombin antibodies. Twenty-four of 25 patients with an activated partial thromboplastin tim...
متن کاملRole of Interleukin-37 in Inflammatory and Autoimmune Diseases
Interleukin-1 family 7 (IL-1F7) is a novel member of IL-1F cytokines. IL-1F7 is more commonly known as IL-37. IL-37 join the α-subunit of the IL-18 receptor, or IL-18 binding protein (IL-18BP), and binding of these proteins can enhance the IL-18 suppression. IL-37 also translocates to the cell nucleus and affects gene transcription. IL-37 inhibits the phosphorylation of p38 mitogen-activated pr...
